Product Specific Information
The 6D4 monoclonal antibody reacts with human MHC class I chain-related (MIC) proteins, MICA and MICB. These proteins share 83% amino acid similarity and structural homology with classical HLA molecules but do not bind β2-microglobulin or peptide typical of HLA class I.
MICA and MICB are expressed on endothelial cells, fibroblasts, gastric epithelium, and PHA-stimulated T cells. They act as ligands for NKG2D receptors found on NK cells, γδ T cells, and αβ CD8+ T cells. Human cytomegalovirus (HCMV) can inhibit MICB function to evade NK cell detection. Expression of MICA and MICB has been observed in various epithelial tumors, including breast, lung, ovary, prostate, colon, and kidney.

Applications Tested:
Tested by flow cytometric analysis of HeLa cells. Recommended ≤0.5 µg/test in 100 µL final volume. Optimal titration is advised depending on cell number (10⁵–10⁸ cells/test).

Aggregation: <10% (HPLC)
Filtration: 0.2 µm post-manufacturing filtered
Target Information
MICA encodes a highly polymorphic MHC (HLA) class I chain-related gene A. The protein is expressed on the cell surface but does not associate with β2-microglobulin. MICA functions as a stress-induced antigen recognized by intestinal epithelial γδ T cells.
