Product Specific Information

The 25-D1.16 monoclonal antibody reacts specifically with the ovalbumin-derived peptide SIINFEKL bound to H-2Kb of MHC class I, but not with unbound H-2Kb or irrelevant peptide-bound H-2Kb.
This antibody is useful for tracking the quantity and localization of these specific antigen-presenting cells (APC) in vivo.

Applications Reported: Flow cytometric analysis and immunohistochemical staining.
Applications Tested: Flow cytometric analysis of SIINFEKL-pulsed C57Bl/6 splenocytes (≤0.25 µg/test).
A test is defined as the antibody amount (µg) that stains a cell sample in 100 µL final volume.
Cell number may range from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

SIINFEKL Peptide Pulsing Protocol

1. Add SIINFEKL peptide to cells in flow staining buffer to a final concentration of 30 µM.
2. Incubate cells at 37°C for 2 hours.
3. Wash cells with flow staining buffer.
4. Proceed with cell surface staining as usual.
