Product Specific Information
The Notch family of transmembrane receptors regulates cell-fate decisions during development across species. Upon ligand binding, the Notch receptor is cleaved, and its intracellular domain translocates to the nucleus to suppress lineage-specific genes via transcriptional repressors.
The mN1A antibody specifically reacts with the intracellular domain of mouse and human Notch1, but not with Notch2, 3, or 4. It shows low affinity for the full-length unprocessed forms of Notch1. In mice, Notch mRNA is expressed in hematopoietic cells of the fetal liver, thymus, and bone marrow. In the thymus, Notch1 protein is detected in CD4-CD8- (double-negative) and CD4-CD8+ (single-positive) thymocytes. Studies indicate its role in regulating lymphopoiesis and myelopoiesis.
Applications Reported: Intracellular staining followed by flow cytometric analysis, immunoblotting (WB), and immunohistochemical staining.
Applications Tested: Tested by intracellular staining and flow cytometric analysis of mouse thymocytes. Recommended ≤1 µg per test (100 µL final volume). Optimal antibody titration is advised for best assay performance.
Target Information
The Notch gene family encodes transmembrane proteins with multiple cysteine-rich EGF-like repeats. Four Notch genes (Notch1–4) are known in mammals and are involved in differentiation of the nervous system and other tissues. Delta and Serrate are identified ligands of Notch1. Mature Notch proteins are heterodimeric receptors formed by cleavage of pre-proteins into an extracellular subunit (NEC) and a transmembrane subunit (Ntm) containing the intracellular region.
