Product Specific Information

This DS5MMER monoclonal antibody recognizes mouse MerTK, a 170–210 kDa member of the TAM family of tyrosine kinase receptors (including Axl and Tyro3). MerTK is expressed on tissue macrophages and is involved in the removal of apoptotic cells through ligands Protein S and Gas6, which bind to phosphatidylserine on apoptotic cells. Ligand binding induces MerTK autophosphorylation, activating PI3K and Akt pathways, leading to phagocytosis and inhibition of TLR-induced pro-inflammatory cytokine production.

MerTK deficiency causes autoimmunity, inflammation, and accumulation of apoptotic bodies. MerTK is constitutively released from the cell surface and can be upregulated by LPS stimulation. It is useful to discriminate macrophages from dendritic cells and is often expressed on malignant cells, possibly contributing to immune evasion.

The DS5MMER antibody is effective on paraformaldehyde-fixed cells and inhibits Gas6-induced MerTK phosphorylation (IC50 < 5 nM). It performs well in western blotting, immunoprecipitation, and immunohistochemistry.

Target Information

MERTK (cMER) is a tyrosine kinase proto-oncogene involved in retinal pigment epithelium (RPE) phagocytosis and multiple physiological processes such as cell survival, migration, differentiation, and efferocytosis. Ligand-induced activation leads to autophosphorylation and downstream signaling via GRB2, PLCG2, MAPK1/2, FAK/PTK2, and RAC1.

MERTK plays key roles in macrophage clearance of apoptotic cells, platelet aggregation, cytoskeleton reorganization, and inhibition of TLR-mediated responses through STAT1 activation and induction of SOCS1/3.
