Product Specific Information
1 µg/mL of MA5-45461 was sufficient for detection of Shank3 in 10 µg COS cell lysate transiently transfected with Shank3 by colorimetric immunoblot analysis using goat anti-mouse IgG:HRP as the secondary antibody.
Detects approximately 190 kDa. No cross-reactivity against Shank1 or Shank2.
Target Information
SHANK3 is a major scaffold postsynaptic density protein that interacts with multiple proteins and complexes to orchestrate dendritic spine and synapse formation, maturation, and maintenance. It interconnects NMDA-type and metabotropic glutamate receptors via complexes with GKAP/PSD-95 and HOMER, respectively, and the actin-based cytoskeleton.
SHANK3 plays a role in structural and functional organization of the dendritic spine and synaptic junction through interactions with Arp2/3 and WAVE1 complex, promoting F-actin clustering. It regulates neuronal growth cone motility and NMDA receptor signaling, modulates GRIA1 exocytosis and GRM5/MGLUR5 signaling, and controls AMPA and metabotropic glutamate receptor-mediated synaptic transmission and plasticity.
SHANK3 may be required at early stages of synapse formation and inhibited by IGF1 to promote synapse maturation. (Uniprot)
