Product Specific Information

The APA5 monoclonal antibody reacts with the mouse CD140a molecule, the alpha chain of the platelet-derived growth factor receptor (PDGF receptor). PDGFRα is a receptor tyrosine kinase that forms dimers upon ligand binding and phosphorylates substrates. Dimers may be α/α, β/β, or α/β combinations serving as substrates for kinase activity. CD140a is expressed by embryonic tissues and mesenchymal-derived cells in adult mouse tissues.

Applications Reported:
Used in flow cytometry, Western blotting, and immunohistology of frozen tissue sections. Also reported for blocking ligand binding.

Applications Tested:
Validated by flow cytometric analysis of NIH/3T3 cells at ≤1 µg/test (100 µL final volume). Optimal antibody concentration should be titrated empirically.

Use in sterile environment. 0.2 µm post-manufacturing filtered.

Target Information

PDGFRA is a cell surface receptor tyrosine kinase binding platelet-derived growth factors, mitogens for mesenchymal-origin cells. The bound growth factor determines receptor dimerization (α/α, β/β, or α/β). Knockout mouse studies show PDGFRA is critical for kidney development, with heterozygous mice exhibiting renal defects.
