Product Specific Information
The 9H10 monoclonal antibody reacts with mouse CD152 (CTLA-4), a protein structurally similar to CD28. CTLA-4 is expressed on activated T cells and binds CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28. While CD28 acts as a costimulator, CTLA-4 inhibits IL-2 secretion and cellular proliferation, limiting T-cell activation. The cytoplasmic region of CTLA-4 contains ER retention motifs, leading to intracellular localization upon TCR signaling.

Applications Tested:
The 9H10 antibody was tested by flow cytometric analysis of resting and Con A-stimulated mouse splenocytes. Use ≤0.5 µg per test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Titrate for optimal assay performance.
Additional Notes:
Due to intracellular CTLA-4 localization, detection may require assessment of both surface and intracellular expression.

Target Information
CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor mainly expressed on CD4+ T cells. It binds CD80 and CD86 with higher affinity than CD28. Unlike CD28, which enhances T-cell activation, CTLA4 inhibits T-cell function. Intracellular CTLA4 is also found in regulatory T cells and contributes to their suppressive activity.
