Product Specific Information

The 14D3 monoclonal antibody reacts with human CD152 (CTLA-4), a receptor structurally related to CD28. CTLA-4 is expressed on activated T cells (and sometimes B cells) and binds CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28.
While CD28 acts as a costimulatory molecule, CTLA-4 inhibits T-cell activation by suppressing IL-2 secretion and cell proliferation. The cytoplasmic region of CTLA-4 contains ER retention motifs, leading to intracellular localization of newly synthesized CTLA-4 upon TCR signaling.

The 14D3 antibody also recognizes rhesus monkey CTLA-4 and shows inhibitory activity. It is suitable for flow cytometry and in vitro inhibition assays (Functional Grade recommended for functional assays).

Tested Application: Flow cytometric analysis of PHA-stimulated human PBMCs at ≤0.25 µg/test (100 µL final volume). Optimal titration is recommended.
Due to intracellular localization, detection of both surface and intracellular CTLA-4 may be necessary.

Target Information

CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor mainly expressed on CD4+ T cells. It binds CD80 and CD86 with higher affinity than CD28 but delivers inhibitory signals, suppressing T-cell activation. Intracellular CTLA4 is also found in regulatory T cells and contributes to their immune-regulatory function.
