Target Information
Cyclin D1 (PRAD-1, bcl-1) is a key regulator of the cell cycle, functioning with CDK4 and/or CDK6 to phosphorylate the Rb protein. It is a proto-oncogene often overexpressed in human neoplasms such as mantle cell lymphoma (MCL).
Cyclin D1 promotes protein phosphorylation, mammary epithelial cell proliferation, and fat cell differentiation. The CCND1 gene is located on chromosome 11 and encodes a cytoplasmic and nuclear protein synthesized during the G1 phase.
Cyclin D1 forms complexes with CDK4/CDK6 to drive G1/S transition and regulate CDK inhibitors (p21Cip1, p27Kip1). Without growth factor stimulation, Cyclin D1 is unstable and undergoes ubiquitin-mediated degradation, contributing to G1/S arrest.
As a member of the cyclin family, Cyclin D1 exhibits periodic abundance throughout the cell cycle and interacts with tumor suppressor Rb. Mutations, amplification, or overexpression of Cyclin D1 are frequently associated with tumorigenesis, making it a valuable target for cancer and cell cycle research.
