Product Specific Information

The monoclonal antibody OX108 recognizes human CD200R (OX2), an inhibitory receptor structurally similar to its ligand CD200 but lacking a typical ITIM domain. Instead, CD200R contains an NPXY motif that binds SHIP, inhibiting ERK, JNK, and MAPK p38 pathways, leading to reduced TNFα secretion in activated macrophages. Isoforms of CD200R play significant roles in differentiation, particularly in tolerogenic dendritic cells.

Expression is restricted to hematopoietic cells, including myeloid cells (monocytes, macrophages, DCs, neutrophils, mast cells, basophils), subsets of T lymphocytes, Langerhans cells, and dendritic epidermal T cells. The OX108 epitope is near the CD200 binding site.

Target Information

CD200R (OX2R) is a receptor for the OX-2 membrane glycoprotein. Both receptor and ligand are cell surface glycoproteins with two immunoglobulin-like domains. CD200R is expressed on myeloid lineage cells and functions as a downregulatory signal for myeloid activity. In mice, expression is found on monocytes, neutrophils, T lymphocytes, and mast cells. Alternative splicing generates multiple transcript variants. Dysregulation of CD200R is associated with primary hypertrophic osteoarthropathy.
