Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by a majority of thymocytes, a subpopulation of mature T helper cells, and, in low levels, monocytes. CD4 is a receptor for the human immunodeficiency virus (HIV). SK3 blocks HIV binding and mixed lymphocyte reaction. The SK3 and RPA-T4 monoclonal antibodies do not cross-block binding, suggesting recognition of distinct epitopes.

Applications Reported:
This SK3 (SK-3) antibody has been reported for use in flow cytometric analysis.

Applications Tested:
This antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test. A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL (typically 10⁵–10⁸ cells/test).

Fluorophore Information:
eFluor® 450 is an alternative to Pacific Blue®. Emission at 445 nm and excitation with Violet laser (405 nm). Ensure instrument compatibility.

Target Information

CD4 antigen is involved in recognition of MHC class II molecules and acts as a co-receptor for HIV. It is primarily expressed in T helper cells, and also in monocytes, macrophages, and dendritic cells.
At the tissue level, CD4 expression is found in thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut.
CD4 functions to initiate or augment early T-cell activation via interaction with the T-cell receptor complex and Lck kinase. It may also mediate neuronal damage in infectious and immune-related CNS diseases.
Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
