Product Specific Information

The RM4-5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by a majority of thymocytes, subpopulations of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and plays a crucial role in T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of RM4-5 is blocked by GK1.5.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.125 µg/test (100 µL reaction volume). Cell number range: 10⁵–10⁸ cells/test. Optimize dilution for best performance.

Fluorophore Information:
eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emits at 445 nm (Violet laser compatible). Ensure instrument compatibility.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD4 antigen is a co-receptor for MHC class II molecule recognition and HIV binding. It is mainly expressed on T helper cells, but also found on monocytes, macrophages, and dendritic cells. CD4 expression is detected in thymus, lymph nodes, tonsils, spleen, and some non-lymphoid tissues. It plays a key role in T-cell activation through association with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in immune-related CNS diseases.
