Product Specific Information

The OKT4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface glycoprotein expressed by the majority of thymocytes, a subpopulation of mature T cells (T-helper cells), and at low levels on monocytes. CD4 is a receptor for the human immunodeficiency virus (HIV). The OKT4 antibody recognizes a different epitope than the RPA-T4 monoclonal antibody, and these antibodies do not cross-block each other.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test.
A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL (10⁵–10⁸ cells/test).

Fluorophore Information:
eFluor® 450 is an alternative to Pacific Blue®, emitting at 445 nm and excited at 405 nm (Violet laser). Ensure the instrument supports this fluorochrome.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

The CD4 antigen is involved in the recognition of MHC class II molecules and functions as a co-receptor for HIV. It is primarily expressed on T-helper lymphocytes and may also be found on monocytes, macrophages, and dendritic cells. CD4 expression is observed in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues such as brain and gut. It plays a key role in initiating T-cell activation through interaction with the T-cell receptor complex and protein tyrosine kinase Lck. CD4 may also contribute to neuronal damage in immune-mediated CNS diseases.
