Product Specific Information

The 1C10 monoclonal antibody reacts with mouse CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a member of the TNFR family and is expressed by mouse B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype switching and, together with IL-4, prevents apoptosis and promotes proliferation.

The 1C10 antibody exhibits agonistic activity both in vitro and in vivo.

• Applications Tested: Tested on mouse splenocytes at ≤0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimal titration recommended.

Fluorochrome Information:
Super Bright 436 is excited by the violet laser (405 nm) and emits at 436 nm. Recommended detection filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

CD40 is a single-chain glycoprotein belonging to the TNF/NGF receptor superfamily, homologous to CD30. It is expressed on B cells (except plasma cells), certain epithelial cells, carcinomas, and dendritic cells. CD40 is essential for immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.

CD40 dysfunction is associated with Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency. It also plays a role in amyloid-beta-induced microglial activation, relevant to Alzheimer’s disease pathogenesis.
