Target Information
ERN1 (endoplasmic reticulum to nucleus signaling 1) is a serine/threonine kinase and endoribonuclease, a human homologue of the yeast Ire1 gene product. It plays a key role in ER-based stress signaling by sensing unfolded proteins in the ER and transmitting the signal to the nucleus through splicing of XBP1 mRNA. This process activates the unfolded protein response transcriptional pathway. ERN1 controls IRE1 proteolysis in mammalian cells and is essential for fetal viability. Under stress conditions that lead to unfolded protein accumulation, depletion of IRE1 results in apoptosis.
