Target Information
ERN1 (endoplasmic reticulum to nucleus signaling 1) encodes a serine/threonine kinase that acts as the sensor of unfolded proteins in the ER. It initiates the unfolded protein response by splicing XBP1 mRNA, converting it into a potent transcriptional activator.
IRE1 is essential for viability under stress conditions that cause unfolded protein accumulation in the ER. Depletion of IRE1p via dominant negative expression leads to apoptosis under such stress conditions.
ERN1 plays a critical role in extraembryonic cells necessary for fetal viability and controls IRE1 proteolysis in mammalian cells.
