Product Specific Information

The monoclonal antibody 2D3 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five transmembrane protein involved in adhesion through interactions with SIRP and integrins (CD51/CD61, CD41/CD61). It mediates bi-directional signaling affecting neural synaptic activity and macrophage phagocytosis. CD47 is also a receptor for thrombospondin and is expressed broadly in hematopoietic and non-hematopoietic cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.6 µg) per test.
A test is defined as the amount of antibody staining a cell sample in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Based on Phiton™ technology using nucleic acid dye structures engineered for minimal spillover and spread impact.

Target Information

CD47 (integrin-associated protein, IAP) is a ubiquitously expressed cell surface transmembrane glycoprotein interacting with integrins and regulating signaling pathways. It modulates intracellular calcium upon adhesion, serves as a receptor for thrombospondin, and influences membrane transport and signal transduction. CD47 interacts with CD172a (SIRPα) on macrophages, preventing phagocytosis of CD47-positive cells. It also affects cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, being abundant in synapse-rich regions of brain and retina.
