Product Specific Information

The monoclonal antibody 2D3 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein with a small alternatively spliced cytoplasmic domain. It interacts with SIRP (signal regulator protein) and β3 integrins (CD51/CD61 and CD41/CD61), mediating bi-directional signaling that affects neural synaptic activity and macrophage phagocytosis. CD47 also serves as a receptor for thrombospondin.

Expression is found in most hematopoietic cells (T cells, B cells, monocytes, platelets, erythrocytes) and in non-hematopoietic tissues.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
Recommended use: 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
Add 5 µL per sample to antibody cocktails or single-color controls.

Target Information

CD47 (integrin-associated protein, IAP) is a ubiquitously expressed transmembrane glycoprotein that interacts with integrins and modulates signal transduction pathways, including activation of G proteins and calcium flux during cell adhesion. It acts as a receptor for thrombospondin and ligand for CD172a (SIRPα), inhibiting macrophage phagocytosis of CD47-positive cells. CD47 also influences cell migration, adhesion, T-cell activation, chondrocyte mechanotransduction, and neuronal development, being abundant in synapse-rich brain and retinal regions.
