Product Specific Information

The monoclonal antibody 2D3 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein involved in adhesion through interactions with SIRP and integrins CD51/CD61 and CD41/CD61. It mediates bi-directional signaling that modifies neural synaptic activity and regulates macrophage phagocytosis. CD47 acts as a receptor for thrombospondin and is expressed in most hematopoietic and non-hematopoietic cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Recommended Usage:

• Use 5 µL (0.6 µg) per test (100 µL final volume)
• Cell count range: 10⁵–10⁸ cells/test
• Add 5 µL of CellBlox Plus Blocking Buffer per sample for optimal staining

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI)
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining
• For single-color controls, use 5 µL of buffer per 100 µL cell sample

Target Information

CD47 (integrin-associated protein, IAP) is a ubiquitously expressed transmembrane glycoprotein that interacts with integrins and modulates signaling pathways, including G protein activation and calcium flux upon adhesion. It serves as a receptor for thrombospondin and plays a role in membrane transport and signal transduction. CD47 binds CD172a (SIRPα) on macrophages, inhibiting phagocytosis of CD47-positive cells. It is involved in cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, being abundant in synapse-rich brain and retinal regions.
