TNFR1 (Tumor necrosis factor receptor 1)는 TNF 슈퍼패밀리에 속하며 주요 TNF-α 수용체 중 하나입니다. TNFR1은 사이토카인 매개 신호전달, NF-κB 경로의 양성 조절, 혈관신생의 양성 조절, 유전자 발현의 음성 조절 등에 관여합니다. TNFR1의 세포외 도메인은 용해성 형태(sTNFR1)로 순환계에 방출됩니다. 인간에서는 TNFR1 유전자가 12번 염색체에 위치합니다. BCL2-associated athanogene 4 (BAG4/SODD), TRADD, TRAF2 단백질과의 상호작용을 통해 TNFR1 신호전달이 조절됩니다. TNFR1의 세포외 도메인 돌연변이는 상염색체 우성 열성 발열 증후군과 관련이 있으며, 수용체 제거 장애가 질병의 원인으로 제시됩니다.
