Product Specific Information

The monoclonal antibody OX104 recognizes human CD200 (OX2), a member of the Ig superfamily with two Ig domains, transmembrane and cytoplasmic regions. CD200 is expressed on resting and activated B and T cells, keratinocytes, nerve cells, dendritic cells, and ovarian cells. Interaction with CD200R modulates immune responses by inhibiting macrophage activation and mast cell degranulation, reducing TNFα and IL-13 secretion, and promoting lymphocyte tolerance. Expression on trophoblasts suggests a role in maternal tolerance.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Usage Recommendations:

• Add 5 µL of CellBlox Plus Blocking Buffer per sample (100 µL cell suspension).
• For single-color controls, use 5 µL per 100 µL sample containing 10³–10⁸ cells.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

Target Information

CD200 is a type I membrane glycoprotein with two immunoglobulin domains, part of the immunoglobulin superfamily. It interacts with CD200R on myeloid cells to inhibit macrophage activation. Homologs are found in herpesviruses and poxviruses. CD200 may regulate myeloid cell activity and deliver inhibitory signals in various tissues.
Associated diseases include Mantle Cell Lymphoma and Primary Hypertrophic Osteoarthropathy.
Multiple alternatively spliced transcript variants encode different isoforms of CD200.
