Product Specific Information

The monoclonal antibody OX104 recognizes human CD200 (OX2), a member of the immunoglobulin superfamily with two Ig domains, a transmembrane, and cytoplasmic domain. CD200 is expressed on resting and activated B cells, subsets of T cells, keratinocytes, peripheral and central nerve cells, follicular dendritic cells, and ovarian cells.

Interaction with CD200R results in:

• Macrophage activation (IL-6 production)
• Inhibition of mast cell degranulation
• Reduction of TNFα and IL-13 secretion
• Attenuation of lymphocyte activation

CD200 expression on trophoblasts suggests a role in maternal tolerance.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on normal human peripheral blood cells. Recommended ≤0.06 µg/test (100 µL final volume). Optimal titration required.

Note:

• NovaFluor dyes are not compatible with DNA intercalating dyes such as propidium iodide, 7-AAD, or DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample or per 100 µL of cell sample (10³–10⁸ cells).
  • Add buffer to antibody cocktails before combining with cells.

Target Information

CD200 is a type I membrane glycoprotein with two immunoglobulin domains, belonging to the immunoglobulin superfamily. It interacts with CD200R on myeloid cells to inhibit macrophage activation. Homologues exist in herpesviruses and poxviruses. CD200 regulates myeloid cell activity and provides inhibitory signals in various tissues. Multiple alternatively spliced variants have been identified.
