Product Specific Information

Description:
Monoclonal antibody OX104 recognizes human CD200 (OX2), a member of the Ig superfamily with two Ig domains, a transmembrane and cytoplasmic domain. CD200 is expressed on resting and activated B cells, subsets of T cells, keratinocytes, peripheral and central nerve cells, follicular dendritic cells, and ovarian cells. Interaction with CD200R affects macrophage activation, mast cell degranulation, and cytokine secretion, contributing to immune regulation and maternal tolerance.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤0.06 µg per test in 100 µL final volume. Cell number should be empirically determined (10⁵–10⁸ cells/test). Titrate antibody for optimal performance.

Compatibility:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

CellBlox Plus Blocking Buffer:
This product ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01). Add 5 µL per sample prior to combining with cells. For single-color controls, add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD200 is a type-1 membrane glycoprotein with two immunoglobulin domains, part of the immunoglobulin superfamily. It interacts with CD200R on myeloid cells to inhibit macrophage activation. Homologues exist in herpesviruses and poxviruses. CD200 may regulate myeloid activity and inhibit macrophage lineage activation. Multiple splice variants exist. Associated diseases include Mantle Cell Lymphoma and Primary Hypertrophic Osteoarthropathy.
