Product Specific Information

The monoclonal antibody OX104 recognizes human CD200 (OX2), a member of the immunoglobulin superfamily with two Ig domains, a transmembrane, and a cytoplasmic domain. CD200 is expressed on resting and activated B cells, subsets of T cells, keratinocytes, nerve cells, follicular dendritic cells, and ovarian cells. Interaction with CD200R modulates immune responses by attenuating macrophage activation and cytokine secretion.

Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

• Applications Reported: Suitable for flow cytometric analysis.
• Applications Tested: Tested on normal human peripheral blood cells. Recommended ≤0.06 µg/test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test.
• Optimization: Titrate antibody for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample, regardless of the number of NovaFluor conjugates.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, utilizing engineered nucleic acid dye structures for improved spectral performance and minimal spillover.

Target Information

CD200 is a type-1 membrane glycoprotein with two immunoglobulin domains, belonging to the Ig superfamily. It interacts with CD200R on myeloid cells to inhibit macrophage activation. CD200 homologues are found in several viruses. Dysregulation is associated with diseases such as Mantle Cell Lymphoma and Primary Hypertrophic Osteoarthropathy. Multiple alternatively spliced transcript variants exist.
