Product Specific Information

The monoclonal antibody OX104 recognizes human CD200 (OX2), a member of the Ig superfamily with two Ig domains, a transmembrane, and a cytoplasmic domain. CD200 is expressed on resting and activated B cells, subsets of T cells, keratinocytes, neuronal cells, dendritic cells, and ovarian cells. Interaction with CD200R modulates macrophage activation, inhibits mast cell degranulation, and attenuates lymphocyte activation. CD200 expression on trophoblasts suggests a role in maternal tolerance.

Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

• Applications Tested: Tested on normal human peripheral blood cells. Recommended ≤0.06 µg/test (in 100 µL volume, 10⁵–10⁸ cells/test).
• Optimization: Antibody titration recommended for best performance.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.

CellBlox Plus Blocking Buffer Usage:

• Add 5 µL per sample (regardless of number of NovaFluors).
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).
• Always add blocking buffer before combining antibody cocktails with cells.

Technology:
NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures with minimal spillover and spread.

Target Information

CD200 is a type I membrane glycoprotein with two immunoglobulin domains, belonging to the Ig superfamily. It interacts with CD200R on myeloid cells to inhibit macrophage activation. Homologs exist in herpesviruses and poxviruses. CD200 regulates myeloid activity and provides inhibitory signals for macrophage lineages. Multiple alternatively spliced variants exist.
