Product Specific Information

The monoclonal antibody B6H12 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein involved in adhesion through interactions with SIRP and integrins (CD51/CD61, CD41/CD61). It mediates bidirectional signaling, regulates macrophage phagocytosis, and acts as a receptor for thrombospondin.

CD47 expression is found in most hematopoietic cells (T/B cells, monocytes, platelets, erythrocytes) and various non-hematopoietic cells. The antibody has reported neutralizing activity.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis and immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections.
Applications Tested: Pre-titrated for flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.8 µg) per test (approx. 10⁵–10⁸ cells/test).

NovaFluor Compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample or per 100 µL of cell suspension.

Technology:
NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD47 (Integrin-associated protein, IAP) is a transmembrane glycoprotein interacting with integrins and regulating their functions. It modulates signaling pathways and intracellular calcium levels upon adhesion. CD47 acts as a receptor for thrombospondin and ligand for CD172a (SIRPα), preventing phagocytosis of CD47-positive cells. It influences immune cell adhesion, migration, activation, and neuronal development, being abundant in synapse-rich regions of the brain and retina.
