The monoclonal antibody PAJ-17R recognizes mouse IL-17 receptor A (IL-17RA), also known as CD217.
IL-17RA is a single transmembrane receptor that can homo- or heterodimerize with IL-17RC or IL-17RB and binds to IL-17A, IL-17F, or IL-17E (IL-25). Ligand binding induces receptor dimerization, recruitment of Act-1 via SEFIR domains, and activation of NFκB and MAPK pathways.

Although early studies suggested ubiquitous expression of IL-17RA, not all IL-17RA-expressing cells respond to IL-17A. Knock-out mouse studies show IL-17RA’s crucial role in neutrophil development and function, with expression found in Gr-1^dim and Gr-1^bright bone marrow cells, as well as some F4/80 positive cells. Staining with PAJ-17R is absent in IL-17RA-deficient mice.

Interleukin 17A (IL-17A) is a proinflammatory cytokine secreted by activated T-lymphocytes and induces the maturation of CD34⁺ hematopoietic precursors into neutrophils.
IL-17RA is a type I membrane glycoprotein that binds IL-17A and IL-17F with low affinity. The IL-17A/IL-17RA axis plays a key role in inflammatory and autoimmune diseases such as rheumatoid arthritis.
Defects in IL-17RA cause familial candidiasis type 5 (CANDF5), characterized by impaired immune responses and reduced fungal clearance.
