Product Specific Information
The eBio64DEC17 antibody reacts with human IL-17A and functions as a neutralizing antibody. IL-17A is a CD4+ T cell-derived cytokine that promotes inflammatory responses and is elevated in conditions such as rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.
Human IL-17A exists as glycosylated 20–30 kDa homodimers produced by activated CD4+ T-cells. It enhances ICAM-1 expression in fibroblasts and stimulates epithelial, endothelial, and fibroblastic cells to secrete IL-6, IL-8, G-CSF, and PGE2. In the presence of IL-17A, fibroblasts support CD34+ progenitor proliferation and neutrophil maturation.
IL-23-dependent, IL-17A-producing CD4+ T cells (Th17 cells) represent a distinct subset from Th1 and Th2 cells. IFN-γ and IL-4 negatively regulate Th17 differentiation.
Intracellular staining by eBio64DEC17 identifies the same population as eBio64CAP17 in co-staining experiments.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated for intracellular staining and flow cytometry of stimulated human peripheral blood cells.
Recommended usage: 5 µL (0.06 µg) per test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test.
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Interleukin-17A (IL-17A, CTLA-8) is a 32 kDa disulfide-linked homodimer of 136 amino acids, a member of the IL-17 family (A–F). It signals through IL-17R (CDw217) and regulates NF-κB and MAPK pathways, stimulating IL-6 and COX-2 expression and nitric oxide production. IL-17A plays key roles in inflammatory and autoimmune responses.
