Product Specific Information

The V3NTY24 monoclonal antibody recognizes human and mouse eukaryotic translation initiation factor eIF4E-binding protein 1 (4E-BP1) when phosphorylated at threonine 37 and/or threonine 46.
4E-BP1 binds to eIF4E and represses cap-dependent translation in its non-phosphorylated form. Phosphorylation at multiple sites disrupts this interaction, enabling translation.
Kinases such as FRAP/mTOR phosphorylate Thr37 and Thr46, while ATM phosphorylates Ser111. PI3 kinase inhibitors LY294002 and wortmannin inhibit phosphorylation at Thr37/Thr46.

Applications Reported:

  • Intracellular staining followed by flow cytometric analysis.

Applications Tested:

  • Pre-titrated and validated for intracellular staining and flow cytometry of stimulated normal human peripheral blood cells.
  • Recommended usage: 5 µL (0.06 µg) per test in 100 µL final volume (10⁵–10⁸ cells/test).

Protocols:

  • Protocol A: Two-step protocol for intracellular (cytoplasmic) proteins.
  • Protocol C: Two-step protocol with fixation/methanol for phospho-specific discrimination.
  • Protocol B: One-step nuclear protein staining not recommended.

Target Information

This gene encodes a translation repressor protein that interacts with eukaryotic translation initiation factor 4E (eIF4E), inhibiting complex assembly and repressing translation.
Phosphorylation in response to stimuli such as UV irradiation or insulin signaling dissociates it from eIF4E, activating mRNA translation.
