Target Information
Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of plasma proteins. It inhibits amyloid fibril formation by several proteins (e.g., APP, APOC2, B2M, CALCA, CSN3, SNCA, and LYZ variants) without requiring ATP. Clusterin maintains partially unfolded proteins for refolding by other chaperones (e.g., HSPA8/HSC70) but does not refold them itself.
Binding to cell surface receptors triggers internalization and degradation of the chaperone–client complex. Secreted isoform 1 protects cells against apoptosis and complement-mediated cytolysis. Intracellular isoforms interact with ubiquitin and E3 ligase complexes, promoting proteasomal degradation of target proteins such as COMMD1 and IKBKB, and modulate NF-κB transcriptional activity. Nuclear isoforms promote apoptosis, while mitochondrial isoforms inhibit BAX-dependent cytochrome c release and apoptosis. Clusterin plays a key role in regulating cell proliferation.
