Target Information

Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of blood plasma proteins. It inhibits amyloid fibril formation by APP, APOC2, B2M, CALCA, CSN3, SNCA, and aggregation-prone LYZ variants (in vitro). Clusterin maintains partially unfolded proteins for refolding by other chaperones such as HSPA8/HSC70 but does not refold proteins itself. Binding to cell surface receptors triggers internalization and subsequent lysosomal or proteasomal degradation. Secreted isoforms protect against apoptosis and complement-mediated cytolysis, while intracellular forms promote ubiquitination and degradation of target proteins, modulating NF-kappa-B activity. Nuclear isoforms promote apoptosis; mitochondrial isoforms inhibit BAX-dependent cytochrome c release, suppressing apoptosis. Plays a role in cell proliferation regulation.
