    Clusterin (also known as complement lysis inhibitor (CLI), apolipoprotein J (APOJ), sulfated glycoprotein 2 (SGP2), SP40, and testosterone-repressed prostate message 2 (TRPM2)) is a secretory, heterodimeric glycoprotein involved in immune regulation, cell adhesion, transformation, lipid transport, tissue remodeling, and membrane recycling. It is synthesized as a 449 amino acid polypeptide that is post-translationally cleaved between Arg227 and Ser228. Clusterin has been associated with amyloid structure formation and may influence Alzheimer’s disease pathogenesis.

Target Information

Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and amyloid fibril formation. It maintains proteins in a state suitable for refolding by other chaperones (e.g., HSPA8/HSC70) but does not refold proteins by itself. Binding to cell surface receptors triggers internalization and degradation of complexes. Secreted isoform 1 protects cells against apoptosis and complement cytolysis, while intracellular isoforms interact with ubiquitin ligase complexes to promote proteasomal degradation of target proteins such as COMMD1 and IKBKB. Clusterin modulates NF-κB activity, influences apoptosis, and plays roles in cell proliferation regulation.
