Product Specific Information

The eBioMWReg30 monoclonal antibody reacts with mouse CD41 (fibrinogen receptor, gpIIb, integrin alpha IIb). CD41 is expressed on platelets, megakaryocytes, and hematopoietic progenitors in embryos and adults. It associates with CD61 (gpIIIa, integrin beta III) to form a receptor essential for platelet adhesion and aggregation, binding to molecules such as fibrinogen, fibronectin, and vitronectin.

CD41 expression can be used with CD48 and CD150 markers to identify hematopoietic stem cells (HSCs). CD150⁺CD48⁻CD41⁻ cells are highly efficient in long-term multilineage reconstitution.

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01)

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample (regardless of number of NovaFluors in panel)
• For single-color controls: use 5 µL per 100 µL of cell sample (10³–10⁸ cells)

Target Information

CD41 (platelet glycoprotein IIb, ITGA2B) consists of alpha (120 kDa) and beta (23 kDa) subunits that interact with CD61 in the presence of calcium to form a functional adhesive receptor. It mediates platelet aggregation by binding to von Willebrand factor, fibrinogen, fibronectin, and vitronectin. CD41 is expressed on megakaryocyte-platelet lineage and early hematopoietic cells.
Associated diseases include Glanzmann Thrombasthenia and Platelet type-16 bleeding disorder.
