Product Specific Information
The monoclonal antibody Moushi101 recognizes mouse CD101, also known as IGSF2 and V7 (7 IgV domains). CD101 is a transmembrane glycoprotein containing 7 Ig-like IgV domains and a short cytoplasmic tail with several phosphorylation sites. Cross-linking CD101 can antagonize TCR/CD3-mediated lymphocyte activation. CD101 is expressed on monocytes, granulocytes, dendritic cells, Langerhans cells, and activated T cells. It is also found on a subset of CD25+Foxp3+ T regulatory cells with high suppressive properties in vitro and in vivo.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.25 µg per test (100 µL final volume). Cell number can range from 10⁵ to 10⁸ cells/test. Antibody titration advised for optimal performance.
Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.
CellBlox Plus Blocking Buffer Usage:
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample (regardless of the number of NovaFluors in the panel). For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).
NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered for optimized spectral performance and minimal spillover.

Target Information
CD101 (V7 antigen) is a member of the immunoglobulin superfamily and forms a disulfide-linked homodimer. It is expressed on monocytes, granulocytes, dendritic cells, and subsets of T lymphocytes. CD101+ CD28+ cells are highly responsive to CD28 signaling, and co-stimulation with anti-CD28 or suboptimal anti-CD3 enhances proliferation, indicating an activating role. The BB27 monoclonal antibody inhibits T cell reactivity in mixed DC–T cell cultures. Mouse CD101 is expressed on CD4+CD25+Foxp3+ regulatory T cells with high suppressor activity. In humans, CD101 is found on ~30% of Foxp3+ PBMCs, though higher suppressor activity has not been confirmed.
