Product Specific Information
- For immunohistochemistry (PFA-fixed): Incubate antibody for 30 minutes at room temperature.
- For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, then cool at room temperature for 20 minutes before staining.
Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide deprivation. p53 activation leads to transcription of genes such as p21waf1, regulating G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.
Phosphorylation at Ser15 is crucial for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells, contributing to anticancer mechanisms. It is also linked to aging, oxidative stress, and cognitive processes such as long-term potentiation (LTP). In normal cells, p53 levels are low, but they are elevated in transformed cell lines, contributing to malignancy. Mutations in the TP53 gene can lead to loss of tumor suppressor function and are implicated in various human cancers, including Li-Fraumeni syndrome.
