Product Specific Information

  • For immunohistochemistry (PFA fixed), incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 upregulates genes such as p21^waf1^, leading to G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation at Ser15 is key for activation and stabilization, while Ser392 phosphorylation is associated with tumor formation. p53 functions as a guardian against proliferation of damaged cells and acts as an anticancer factor. It is found at low levels in normal cells but elevated in transformed or malignant cells. Mutations in TP53 often cause loss of DNA binding and tumor suppressor activity, contributing to cancer development. Germline TP53 mutations are associated with cancer-prone syndromes such as Li-Fraumeni syndrome.
