Product Specific Information
- For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 activation leads to transcription of genes such as p21waf1, contributing to G1 or G2/M arrest.
Structurally, p53 comprises:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerization domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is crucial for p53 activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays key roles in suppressing proliferation of damaged cells, aging, oxidative stress responses, and cognitive functions such as long-term potentiation (LTP).
In normal cells, p53 is expressed at low levels but is upregulated in transformed cell lines, contributing to malignancy. Mutations in the TP53 gene lead to loss of tumor suppressor activity and are common in many human cancers. Germline mutations are also found in cancer-prone families, such as those with Li-Fraumeni syndrome.
