Product Specific Information

  • This antibody does not react with Mouse or Rat.
  • For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 upregulation leads to activation of genes such as p21waf1, involved in G1 or G2/M arrest. Its structure includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation at Ser15 is crucial for p53 activation and stabilization, while phosphorylation at Ser392 is associated with human tumor formation. p53 plays a role in preventing proliferation of damaged cells and acts as an anticancer agent. It is also linked to aging, oxidative stress, and cognitive processes such as long-term potentiation (LTP). While expressed at low levels in normal cells, p53 is overexpressed in many transformed cell lines, contributing to malignancy. Mutations in TP53 result in loss of tumor suppressor activity and are frequently found in human cancers, including germline mutations in Li-Fraumeni syndrome.
