Product Specific Information

Description:
The OKT4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface glycoprotein expressed by most thymocytes, a subpopulation of mature T-helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV. The OKT4 antibody recognizes a distinct epitope from the RPA-T4 antibody and does not cross-block binding.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested on normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test in a final volume of 100 µL. Cell count range: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emits at 702 nm. Recommended filter: 710/50 bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in:

• IC Fixation Buffer (Product # 00-8222)
• 1-step Fix/Lyse Solution (Product # 00-5333)
  for up to 3 days at 4°C in the dark with minimal brightness loss.

Target Information

The CD4 antigen participates in the recognition of MHC class II molecules and serves as a co-receptor for HIV. It is primarily expressed on T-helper lymphocytes and also found on monocytes, macrophages, and dendritic cells. CD4 expression occurs in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues such as brain and gut. CD4 aids T-cell activation via association with the T-cell receptor complex and tyrosine kinase Lck, and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
