Product Specific Information

The 17A2 monoclonal antibody reacts with the mouse CD3 complex. CD3 subunits (gamma, delta, epsilon) are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells. Binding of 17A2 to CD3 initiates intracellular signaling leading to activation and proliferation.

• Applications Reported: Flow cytometric analysis and functional activity (in vitro stimulation)
• Applications Tested: Tested on mouse thymocytes and splenocytes. Recommended ≤1.0 µg/test (100 µL final volume). Optimize empirically for best performance.
• Fluorochrome: Super Bright 702, excited at 405 nm and emits at 702 nm (recommended 710/50 bandpass filter).
• Buffer Recommendation: Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright dyes to prevent non-specific polymer interactions.
• Light Sensitivity: Protect from light to avoid photo-induced oxidation.
• Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark. Clone-specific performance should be verified empirically.
• Laser Compatibility: Violet laser (405 nm excitation, 702 nm emission).

Target Information

The CD3 complex is essential for signal transduction in T cells and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded on chromosome 11. These subunits contain cytoplasmic tails with tyrosine-based motifs that interact with signal transduction molecules. CD3 is expressed on most T cells, thymocytes, and Purkinje cells, but not on B or NK cells. Defects in CD3 genes can lead to immunodeficiency.
