Product Specific Information

The RPA-T4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface receptor expressed by most thymocytes, a subpopulation of mature T cells (T-helper cells), and at low levels on monocytes. CD4 acts as a receptor for the human immunodeficiency virus (HIV). RPA-T4 blocks HIV binding and mixed lymphocyte reaction. This antibody recognizes a different epitope than the OKT4 monoclonal antibody and does not cross-block binding.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test (final volume 100 µL). Cell number: 10⁵–10⁸ cells/test (empirically determined).

Fluorochrome Information:
Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emits at 702 nm. Recommended filter: 710/50 bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark. Clone-specific performance should be verified empirically.

Target Information

CD4 antigen is involved in MHC class II recognition and serves as a co-receptor for HIV. It is primarily expressed in T-helper lymphocytes, as well as monocytes, macrophages, and dendritic cells. CD4 expression is observed in thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut. It plays a key role in early T-cell activation via association with the T-cell receptor complex and Lck kinase. CD4 may also mediate neuronal damage in infectious or immune-mediated CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
