Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta subunits, is essential for assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells.

Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling that leads to activation and proliferation.

Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.25 µg)/test (100 µL final volume, 10⁵–10⁸ cells/test)

Fluorochrome Information:
Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emits at 702 nm. Recommended filter: 710/50 bandpass.

Staining Panel Recommendation:
Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright dyes to minimize non-specific interactions.

Light Sensitivity:
Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness.

Target Information

The CD3 complex transduces antigen-recognition signals into T cell cytoplasm and regulates TCR surface expression. It includes gamma, delta, epsilon, and zeta subunits encoded on chromosome 11. CD3 is expressed on most T lymphocytes and thymocytes, but not on B or NK cells. Defects in CD3 genes are linked to CD3 immunodeficiency.
