Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by thymocytes, T helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 recognize distinct epitopes.

Applications Reported:

• Flow cytometric analysis.

Applications Tested:

• Pre-titrated and tested on normal human peripheral blood cells.
• Recommended use: 5 µL (0.06 µg) per test (100 µL final volume).
• Cell number: 10⁵–10⁸ cells/test (empirically determined).

Fluorochrome Information:

• Super Bright 702 is a tandem dye excited at 405 nm and emits at 702 nm.
• Recommended filter: 710/50 bandpass.
• Ensure instrument compatibility.

Staining Recommendations:

• Use Super Bright Complete Staining Buffer (Product # SB-4401) for multiple Super Bright conjugates to minimize polymer interactions.

Light Sensitivity:

• Sensitive to photo-induced oxidation. Protect vial and samples from light.

Fixation:

• Samples can be stored up to 3 days in the dark at 4°C using:
  • IC Fixation Buffer (Product # 00-8222)
  • 1-step Fix/Lyse Solution (Product # 00-5333)
• Clone-specific performance should be empirically determined.

Target Information

The CD4 antigen is a co-receptor for HIV and interacts with MHC class II molecules. It is mainly expressed on T helper cells, but also found on monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut. CD4 aids in T-cell activation via association with Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
