Product Specific Information
The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the Ig superfamily. PD-1 contains the immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease in mice. PD-1 is expressed mainly on activated T and B lymphocytes. PD-L1 (B7-H1) and PD-L2 (B7-DC) are known ligands of PD-1, exhibiting overlapping functions and expression patterns.
J43 inhibits the binding of mouse PD-L1-Ig and PD-L2-Ig to PD-1/BHK transfected cells. In vivo, J43 enhances contact hypersensitivity and exacerbates acute GVHD, EAE, and NOD diabetes, similar to PD-1-deficient cell transfer or antibodies against PD-L1/PD-L2.
Applications Reported: Flow cytometric analysis, immunoprecipitation, and functional assays.
For functional assays, use Functional Grade purified J43 (Product #16-9985).
Applications Tested: Flow cytometry of mouse Con-A activated spleen cells and PD-1 transfected cells at ≤0.5 µg/test (100 µL final volume). Optimize antibody titration per assay.

Target Information
PD-1 (PDCD-1) is an inhibitory receptor expressed on activated T cells. It binds PD-L1 and PD-L2 on antigen-presenting cells to suppress T-cell activation in the absence of danger signals. PD-1 deficiency results in hyperactivated immune responses and autoimmune disease. PD-1 may migrate at higher molecular weight than predicted in SDS-PAGE.
