Product Specific Information
The 2B11 monoclonal antibody reacts with mouse and human CXCR4 (CD184, Fusin, LESTR, HUMSTR). CXCR4 is a G-protein-coupled chemokine receptor that functions as a coreceptor for X4 HIV-1 entry into CD4+ cells and plays a key role in lymphocyte migration. Its ligand is SDF1, and it is essential for granulocyte progenitor maintenance and B cell development.
Expression of CXCR4 differs between species. In mice, CXCR4 is restricted to CD4+CD8+ thymocytes, while in humans it is predominantly expressed on B lymphocytes in the spleen.
The 2B11 antibody recognizes the amino terminus of CXCR4 and induces receptor internalization without blocking SDF1α binding.
Applications include flow cytometry, Western blotting, and frozen tissue immunohistology.
Tested Application: Flow cytometric analysis of mouse thymocytes (≤1 µg/test, 100 µL final volume).
Purity: >90% (SDS-PAGE)
Aggregation: <10% (HPLC)
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CXCR4 (Fusin, LESTR, HUMSTR) belongs to the G-protein-coupled chemokine receptor family with seven transmembrane domains. It acts as a co-receptor for HIV-1 and HIV-2 entry via CD4 binding and mediates Env-induced fusion. It also serves as a receptor for CXCL12/SDF-1, leading to calcium signaling and MAPK activation.
CXCR4 has roles in hematopoiesis, cardiac and vascular development, and neuronal survival. It binds bacterial LPS and mediates inflammatory responses. Antibodies against CXCR4 can block HIV-1 and HIV-2 fusion and infection.
