Product Specific Information

The J116 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the immunoglobulin superfamily. PD-1 contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease. It is expressed predominantly on activated T and B lymphocytes. PD-1 ligands include PD-L1 (B7-H1) and PD-L2 (B7-DC), which exhibit overlapping functions and are expressed on normal and activated antigen-presenting cells.

Binding of the J116 antibody inhibits PD-1 signal transduction but does not block PD-L1 binding.

Applications Tested:
Tested by flow cytometric analysis of human PD-1 transfected cells. Recommended ≤1 µg/test (100 µL final volume). Optimal titration is advised for best performance.

Target Information

T-cell activation is regulated by PD-1 (PDCD-1), which binds ligands PD-L1 and PD-L2 to mediate inhibitory signals. This mechanism maintains immune self-tolerance and prevents overactivation. PD-1 knockout mice show hyperactivated immune systems and autoimmune phenotypes. PD-1 often migrates at higher molecular weights in SDS-PAGE than predicted.
