Product Specific Information
Description:
The TM-b1 monoclonal antibody reacts with mouse CD122, the 90–110 kDa interleukin-2 receptor beta chain (IL-2Rβ). CD122 is expressed by NK cells and some T cells in the periphery, and is upregulated upon activation. CD122 associates with CD25 and CD132 (common γ chain) to form the high-affinity IL-2 receptor. High-affinity IL-2 binding is inhibited by the TM-b1 antibody.

Reported for use in flow cytometric analysis.
Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.25 µg per test (100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test). Optimize antibody titration for best results.
Fluorophore Information:
eFluor™ 450 is an alternative to Pacific Blue™, excited at 405 nm and emitting at 445 nm. Ensure instrument compatibility with this fluorochrome.

Target Information
CD122 (Interleukin 2 receptor) is involved in T cell-mediated immune responses, primarily expressed in the hematopoietic system and present in three forms:
- Low affinity: α subunit monomer, not involved in signal transduction
- Intermediate affinity: α/β heterodimer
- High affinity: α/β/γ heterotrimer
Intermediate and high affinity forms mediate receptor endocytosis and mitogenic signal transduction. The CD122 gene encodes the β subunit, a type I membrane protein.
Associated diseases include oligoarticular juvenile idiopathic arthritis and rheumatoid factor negative juvenile idiopathic arthritis.
