Product Specific Information

The monoclonal antibody TWAJ recognizes mouse and human Gata-3, a member of the Gata family of transcription factors.
Gata-3 is a T cell–specific transcription factor essential for thymic development and Th2 differentiation. It is expressed in the central nervous system, skin, mammary glands, and kidney during embryogenesis. Homozygous knockout of Gata-3 is embryonic lethal, highlighting its essential role in development.

In the thymus, expression is mainly found in CD4 single-positive cells. During Th2 differentiation, Gata-3 binds to the IL-4 promoter and represses T-bet expression, inhibiting Th1 differentiation.
Alternative splice variants (50 and 45 kDa) have been reported, and this antibody recognizes both forms.

  • Buffer Requirement: Use Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) for staining.
  • Cross-reactivity: Published in rhesus monkey.
  • Applications Reported: Intracellular staining followed by flow cytometric analysis.
  • Applications Tested: Pre-diluted and validated for intracellular staining of mouse thymocytes using Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
    Recommended usage: 5 µL (1.0 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Violet™ 480 (BV480) emits at 479 nm and is suitable for cytometers with a violet (405 nm) laser. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using multiple polymer dye–conjugated antibodies (e.g., Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to reduce non-specific interactions.

Fixation Compatibility:
BV480 is not compatible with methanol-based fixation.

Target Information

The GATA3 gene encodes a zinc finger transcription factor that binds enhancer elements of all four T-cell receptor (TCR) genes (alpha, beta, gamma, delta).
It is abundantly expressed in T lymphocytes and plays a key role in T-cell receptor gene activation.
The human gene contains six exons over 17 kb of DNA, with zinc fingers encoded by two conserved exons similar to GATA1.
