Product Specific Information

This 22F6 monoclonal antibody reacts with human, mouse, and canine Helios. Helios is a zinc-finger transcription factor similar to Ikaros and is expressed in hematopoietic stem cells in both embryonic and adult mice. It is highly expressed in regulatory T cells but not in mature B cells, dendritic cells, or myeloid cells. Helios can form homo- or heterodimers with other Ikaros family members to regulate transcription. Eight isoforms exist, with some dominant-negative variants upregulated in lymphoid malignancies, including T-acute lymphoblastic leukemia.

22F6 has been reported to cross-react with rat Helios.

• Intracellular staining followed by flow cytometric analysis.

Applications Tested

• Pre-diluted and tested by intracellular staining of mouse lymph node cells using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
• Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test).

Fluorochrome Information

Brilliant Violet™ 480 (BV480) emits at 479 nm and is suitable for cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility before use.

When using multiple polymer dye-conjugated antibodies (e.g., Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (#SB-4401-42) or Brilliant Stain Buffer™ (#00-4409-75) to minimize non-specific interactions.

Note: BV480 is not compatible with methanol-based fixation.

Target Information

Helios is a member of the Ikaros family of zinc-finger proteins, including Ikaros, Aiolos, and Helios. These hematopoietic-specific transcription factors regulate lymphocyte development. Helios forms homo- or heterodimers with other family members and functions mainly in early hematopoietic development. Multiple transcript variants exist, though not all have confirmed biological validity.
