Product Specific Information

This 13F12F2 monoclonal antibody reacts with human killer cell lectin-like receptor subfamily G, member 1 (KLRG1), a type II transmembrane inhibitory receptor of the C-type lectin superfamily containing an ITIM domain.
It is expressed on subsets of γδ T cells, NK (CD56^dim^), CD8+, and CD4+ T cells, primarily in effector/memory phenotype cells that are short-lived but capable of immediate effector function.

Cadherin-E, -N, and -R serve as ligands for KLRG1, and their interaction inhibits cytolytic activity and proliferation. The percentage of KLRG1⁺ cells varies depending on antigen experience.

Clones 13F12F2 and 13A2 recognize similar epitopes based on cross-blocking studies.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (1.0 µg) per 100 µL cell suspension (10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Violet™ 480 (BV480) emits at 479 nm and is compatible with cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility before use.

When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Brilliant Violet™ 480 is not compatible with methanol-based fixation.

Target Information

KLRG1 (Killer cell lectin-like receptor G1, MAFA, 2F1-Ag) is a homodimeric receptor of the lectin-like type 2 transmembrane family containing ITIM motifs in its cytoplasmic domains. These motifs interact with SH2 domains of phosphatases such as SHP-1.
KLRG1 functions as an inhibitory receptor expressed on NK cells and certain T cells, which are involved in tumor and virus-infected cell lysis and immune regulation.
